Characterization of Pseudomonas syringae pv. morsprunorum originating from sweet cherry and plum in Serbia

چکیده

Pseudomonas strains originating from symptomatic (bacterial spot) leaf tissues of sweet cherry (Topola, Sumadija) and plum (Krusedol Selo, Srem) were isolated during 2016 2020, respectively. Based on the findings yielded by classical microbiological methods, LOPAT (+---+), GATTa (--++) pathogenicity tests performed detached fruitlets (sweet sour cherry) pods (bean pods), all confirmed to belong P. syringae pv. morsprunorum. The detection cfl gene allowed that race 1 be identified. DNA fingerprinting patterns obtained with four rep-PCR (BOX ERIC), RAPD-PCR (M13), IS50-PCR (IS50) methods revealed seven tested s. morsprunorum strains, as well comparative KBNS71 reference strain CFBP 2119, genetically heterogeneous. Conversely, MLSA based four-gene-based scheme (gapA, gltA, gyrB, rpoD) indicated genetic homogeneity among Serbian NCBI. Although indicate used in this study are 100% identical, they might have different virulence genes, genome sequencing should eventually find sub-clades host.

Characterization of alginate lyase from Pseudomonas syringae pv. syringae.

The gene encoding alginate lyase (algL) in Pseudomonas syringae pv. syringae was cloned, sequenced, and overexpressed in Escherichia coli. Alginate lyase activity was optimal when the pH was 7.0 and when assays were conducted at 42 degrees C in the presence of 0.2 M NaCl. In substrate specificity studies, AlgL from P. syringae showed a preference for deacetylated polymannuronic acid. Sequence a...

Effect on lipopolysaccharide structure of aeration during growth of a plum isolate of Pseudomonas syringae pv. morsprunorum.

The composition of lipopolysaccharide (LPS) extracted with aqueous phenol from a virulent English plum isolate of Pseudomonas syringae pv. morsprunorum varied according to the partial pressure of oxygen (pO2) in the culture medium at the time of harvest. When pO2 was low, the organism grew slowly and produced smooth LPS bearing rhamnan sidechains. As pO2 was raised, the rate of growth increased...

Characterization of the hrpZ gene from Pseudomonas syringae pv. maculicolaM2

Pseudomonas syringae pv. maculicola is a natural pathogen of members of the Brassicaceae plant family. Using a transposon-based mutagenesis strategy in Pseudomonas syringaepv. maculicola M2 (PsmM2), we conducted a genetic screen to identify mutants that were capable of growing in M9 medium supplemented with a crude extract from the leaves of Arabidopsis thaliana. A mutant containing a transposo...

Complete Genome Sequence of the Kiwifruit Pathogen Pseudomonas syringae pv. actinidiae Biovar 5, Originating from Japan

We present the first complete genome sequence of a copper-resistant biovar 5 strain of a bacterial pathogen of kiwifruit, Pseudomonas syringae pv. actinidiae. Comparison with the genome sequence of a copper-sensitive biovar 5 isolate indicates that copper resistance is encoded on a plasmid.
